Disconnected (disco)-interacting protein 2 homolog A is a member of the DIP2 protein family encoded by Dip2a gene. Dip2a expression pattern has never been systematically studied. Functions of Dip2a in embryonic development and adult are not known. To investigate Dip2a gene expression and function in embryo and adult, a Dip2a-LacZ mouse model was generated by insertion of β-Gal cDNA after Dip2a promoter using CRISPR/Cas9 technology. Dip2a-LacZ mouse was designed to be a lacZ reporter mouse as well as a Dip2a knockout mouse. Heterozygous mice were used to study endogenous Dip2a expression and homozygotes to study DIP2A-associated structure and function. LacZ staining indicated that Dip2a is broadly expressed in neuronal, reproductive and vascular tissues, as well as in heart, kidney, liver and lung. Results demonstrate that Dip2a is expressed in ectoderm-derived tissues in developing embryos. Adult tissues showed rich staining in neurons, mesenchymal, endothelial, smooth muscle cells and cardiomyocytes by cell types. The expression pattern highly overlaps with FSTL1 and supports previous report that DIP2A to be potential receptor of FSTL1 and its protective roles of cardiomyocytes. Question: What was the role of fire during the establishment of the current overstory (ca. 1870–1940) in mixed‐oak forests of eastern North America? Location: Nine sites representing a 240‐km latitudinal gradient on the Allegheny and Cumberland Plateaus of eastern North America. Methods: Basal cross‐sections were collected from 225 trees. Samples were surfaced, and fire scars were dated. Fire history diagrams were constructed and fire return intervals were calculated for each site. Geographic patterns of fire occurrence, and fire‐climate relationships were assessed. Results: Fire was a frequent and widespread occurrence during the formation of mixed‐oak forests, which initiated after large‐scale land clearing in the region ca. 1870. Fire return ranged from 1.7 to 11.1 years during a period of frequent burning from 1875 to 1936. Fires were widespread during this period, sometimes occurring across the study region in the same year. Fires occurred in a variety of climate conditions, including both drought and non‐drought years. Fires were rare from 1936 to the present. Conclusions: A variety of fire regime characteristics were discerned. First, a period of frequent fire lasted approximately 60 years during the establishment of the current oak overstory. Second, fire occurred during a variety of climate conditions, including wet climates and extreme drought. Finally, there was within‐site temporal variability in fire occurrence. 3-Spiro-(2′-(N-oxyl-4′,4′-dimethyloxazolidine)) — cholestane, (I) and 12-spiro-(2′-(N-oxyl-4′,4′-dimethyloxazolidine))-stearic acid (II) have been used as molecular probes to study the interaction of sphingomyelin and cholesterol in both dry and hydrated oriented films at different temperatures. The presence of 50 mole percent cholesterol causes a gel to liquid crystalline phase transition of bovine brain sphingomyelin at 20°C. A temperature induced phase transition involving the phospholipid polar groups has been detected. The mean transition temperature from a rigid to a fluid bilayer lattice structure is 32°C ±0.5°C in hydrated equimolar sphingomyelin — cholesterol films. 相似文献

The phase behavior of mixtures formed with palmitic acid (PA) and one of the following sterols (dihydrocholesterol, ergosterol, 7-dehydrocholesterol, stigmasterol and stigmastanol), in a PA/sterol molar ratio of 3/7, has been characterized by IR and 2H NMR spectroscopy at different pH. Our study shows that it is possible to form liquid-ordered (lo) lamellar phases with these binary non-phospholipid mixtures. The characterization of alkyl chain dynamics of PA in these systems revealed the large ordering effect of the sterols. It was possible to extrude these systems, using standard extrusion techniques, to form large unilamellar vesicles (LUVs), except in the case of ergosterol-containing mixture. The resulting LUVs displayed a very limited passive permeability consistent with the high sterol concentration. In addition, the stability of these PA/sterol self-assembled bilayers was also found to be pH-sensitive, therefore, potentially useful as nanovectors. By examining different sterols, we could establish some correlations between the structure of these bilayers and their permeability properties. The structure of the side chain at C17 of the sterol appears to play a prime role in the mixing properties with fatty acid. 相似文献

Cytokinin, [8-14C]Benzylaminopurine, metabolism in tomato pericarp was followed during a 3 h period utilizing thin layer chromatography and visualization by fluorography. Fluorography indicated the formation of at least 7 metabolites during 3 h. Cytokinin metabolism was reduced by approximately 40% in 3 h by the presence of 250 microM metyrapone, an inhibitor of cytochrome P-450 related enzyme systems. In the presence of metyrapone, the number of radioactive metabolites on the thin layer plate was reduced from 7 to 4 and 2 of these were unique to the metyrapone-treated sample. These data suggest the initial step in benzylaminopurine metabolism in tomato pericarp may be mediated by a cytochrome P-450 related enzyme system which is altered in the presence of metyrapone. 相似文献
